PagP is an outer membrane enzyme of Gram-negative bacteria that functions to modulate the host immune response to infections. PagP not only fortifies the outer membrane permeability barrier to provide enhanced antimicrobial resistance, but it can also function as a phospholipid-recycling sensory transducer. The co-evolution of eukaryotic hosts with bacteria has revealed multiple roles for PagP to guide the development of vaccine adjuvants and novel strategies to treat infections.
